Question:

Suresh prepares slides with pollen grains in different sugar concentrations (0%, 2.5%, 5%, 7.5%, 10%) to study the germination of pollen.

(A) What are the different hypotheses which can be tested using this set-up?

(B) What parameters should be kept the same in this set-up?

(A) The following hypotheses can be tested using this experimental setup:

(1) Pollen grains germinate better in sugar solutions than in plain water (0% sugar), suggesting that the availability of nutrients in the medium influences germination.

(2) There exists an optimal sugar concentration at which pollen germination rate is the highest; concentrations either lower or higher than this optimum concentration result in reduced germination.

(3) The length of the pollen tube produced increases with increasing sugar concentration up to a certain point, after which it decreases.

(4) Very high sugar concentrations inhibit pollen germination due to osmotic stress (water is drawn out of the pollen grain in a highly concentrated solution).

(5) Pollen grains do not germinate at all in the absence of sugar (0% concentration), indicating that a minimum nutrient concentration is necessary for germination.

(B) To ensure a fair comparison, all conditions except sugar concentration should be kept the same. The same type and age of pollen grains, same number of pollen grains on each slide, same volume of sugar solution, same temperature, same humidity, same time allowed for germination, same slide preparation method, and same microscope magnification should be maintained. This ensures that any difference in pollen germination is due only to the change in sugar concentration.
